Sheath blight, which is caused by Rhizoctonia solani, is a disease that majorly impacts rice production. A biocontrol agent used for control rice sheath blight must be sprayed on the stem at specific times during rice growth, a process that is labour-intensive and renders the antagonist vulnerable to environmental factors. In this study, Trichoderma asperellum T12 was used to produce preparation by solid-state fermentation using a surface-response method. Rice hull was selected as a carrier based on its ability to sustain the T12 floating in the water and protect T12 from ultraviolet irradiation. The production of a T12-based preparation required 32% wheat bran, 7% inoculum, 2.3 g kg?1 (NH4)2SO4 and 65% water content, with fermentation at 27.5°C for 30 days and agitation every six days. The preparation demonstrated 90% biocontrol efficacy and significantly (P > 0.05) increased the seed-set rate and 1000-grain weight as compared with the pathogen treatment. The population of Trichoderma on the surface of rice leaf sheath in the treatment applied with T12 preparation increased from 232 cfu (colony forming units) g?1 fw (fresh weight) to 436 cfu g?1 fw during rice growth stage, which was significantly (P > 0.05) higher than pathogen treatment. The population of R. solani on the leaf sheath increased from 41 cfu g?1 fw to 271 cfu g?1 fw in the pathogen treatment, while remained stable (P > 0.05) at level of 10–23 cfu g?1 fw in T12 preparation applied treatment. Biocontrol of sheath blight by the addition of the preparation to the soil is effective and decreases the costs of agro-industrial waste disposal.  相似文献

Mutations were induced in tissue-cultured wasabi (Wasabia japonica Matsumura) by treating in vitro-derived shoot tips with either γ-rays or X-rays at 0, 10, 20, 40 or 80 Gy. Doses of up to 40 Gy of either γ- or X-ray treatments resulted in a survival rate of more than 60% in culture after 3 mo. The use of γ- or X-rays at doses between 10 Gy and 40 Gy to induce mutation in W. japonica resulted in an alteration of the growth and allyl isothiocyanate (AITC) content of multiple shoots after 3 mo. in culture on Murashige and Skoog medium containing 5 μM N6-benzyladenine (BA). Putative mutants from the 40 Gy treatments of either γ- or X-rays exhibited a reduction in shoot weight, number, and height, whereas treatments of either γ-rays or X-rays at 10 Gy and 20 Gy doses showed no significant differences in shoot growth. All shoots treated with 80 Gy were either necrotic or irregenerable, while those treated with 40 Gy produced deformed leaves, from both types of ionizing radiation. Concentrations of AITC were measured by the use of gas chromatography-mass spectrometry (GC-MS). The accumulation of AITC was shown to decrease when doses increased in both γ- and X-ray treatments, compared with the controls. Positive responses were solely occurred at 18 mo. after transfer of in vitro rooted shoots to the shade house. The survival rate, rhizome weight and AITC content of plants derived from shoots treated with 20 Gy or 40 Gy of either γ-rays or X-rays were significantly greater than those of the controls.  相似文献

Abstract. Fine-scale structure of a species-rich grassland was examined for seasonal changes caused by manipulated changes in the availability of above and below-ground resources (additional illumination with the help of mirrors and fertilization) in a field experiment. If changes induced by fertilization — which are expected to lead to a reduction in small-scale diversity —are due to intensified light competition, they should be compensated for by additional light input. Permanent plots of 40 cm × 40 cm were sampled by the point quadrat method at three angles (60°, 90° and 120° from the horizontal North-South direction), using a laser beam to position the quadrats, in early July and early September. The applied treatments did not cause apparent changes in plant leaf orientation. The degree of spatial aggregation of biomass increased seasonally in fertilized, non-illuminated plots: greater productivity at a constant light supply led to a faster growth rate of potentially dominant species, as compared to the subordinate ones. Additional illumination mitigated this effect of fertilization, indicating that the observed changes in biomass aggregation were due to increased light competition. There was a considerable seasonal decrease of variance ratio (ratio of observed variance of richness at a point and variance expected at random) in fertilized only and in illuminated only plots. In fertilized plots this was due to the positive relationship between biomass aggregation and expected variance of richness. Biomass constancy occurs to be inversely related to deficit in variance of richness. In illuminated plots, in contrast, only the observed variance of richness decreased seasonally, indicating a more uniform use of space by different species. Evidently, a deficit in variance of richness can be caused by drastically different processes, showing that the variance ratio statistic may not have a significant explanatory value in fine-scale community studies.  相似文献

Seven hundred and fifty-two rhizobacteria were isolated from 186 rhizosphere soil samples collected across tomato growing regions of Karnataka. Among them, 26% strains were Gram positive and other 74% were Gram negative and dominant being Bacillus and Pseudomonas. Sampling of different locations showed variation in species richness and diversity indices. Similarity matrix computed with Jaccard’s coefficient and principle coordinate analysis to correlate bacterial diversity revealed that rhizobacterial genera of Mysore, Mandya and Kolar soil samples were very closely related and rarefaction curve analysis indicated that these soil samples also harbored higher number of rhizobacteria which included all the genera studied. PGPR trait analysis revealed that most of the rhizobacteria were endowed with more than one beneficial trait which may act individually or simultaneously, and indole acetic acid production and phosphate solubilization are the two predominant traits exhibited by these rhizobacteria. Rhizobacterial isolates also showed a varied level of plant growth promotion traits and offered protection against fungal origin foliar and root pathogens. Among the nine regions studied, Mysore, Mandya and Kolar regions recorded higher percentage of promising PGPRs in comparison with other regions studied of Karnataka.  相似文献

The development of seedlings of two miombo trees, Brachystegia spiciformis Benth. and Julbernardia paniculata (Benth.) Troupin, was studied during two growing seasons (December 1989–April 1991) at a Zambian grassland site. Seed germination rates under laboratory and field conditions were not significantly different although germination in the field was delayed by 1–2 weeks due to insufficient rainfall. After one year of storage J. paniculata seed germination had declined from 67% to 17% while germination of B. spiciformis seeds remained at about 83%.Leaf production was confined to the rainy season. Leaf fall occurred during the dry season and in J. paniculata this was followed by shoot die-back during the hot dry period (August–November). Two-thirds of B. spiciformis seedlings experienced shoot die-back but shoot die-back did not necessarily result in seedling mortality. Seedling deaths occurred during the germination period (6–10 weeks after planting) and in the hot dry period (40–50 weeks after planting) during September–November. Survivorship of B. spiciformis seedlings was 74% at the end of the second growing season while this was 46% for J. paniculata.Shoot growth was negligible during the second growing season. In fact mean maximum leaf area of B. spiciformis decreased significantly from 19.7 cm2 (SD=5.7) per plant at the end of the first growing season to 13.3 cm2 (SD=5.8) at the end of the second growing season (t=3.31, P<0.01). However, root biomass of B. spiciformis seedlings increased 2.8 times during the second growing season.These results suggest that shoot die-back in seedlings of miombo trees is caused by drought and that the slow shoot growth is the result of allocating most of the biomass to root growth during seedling development.  相似文献

Phytophthora ramorum has been found in waterways outside infested nurseries, but little is known about its behavior in water. This study examined the effect of salinity on survival, growth, sporulation, and infection. P. ramorum survival and growth was negatively correlated with salt concentration (range of 0–45 g l−1), but showed a level of tolerance even at 45 g l−1. No sporangia were observed in cultures with higher than 20 g l−1 of salt and zoospores were not released from sporangia above 14 g l−1. Water sources with different salinity were used to understand the environment where P. ramorum can survive and infect host material. Water from natural bodies and water amended with different salt concentrations were added to P. ramorum-infested sand and baited with rhododendron leaf disks. Infection decreased with increasing salt concentration and increased with higher initial concentration of P. ramorum. This research helps to better understand the effects of water quality on survival and infectivity of P. ramorum, expanding the potential survey range.  相似文献

Bacterial blight induced by Xanthomonas campestris pv. malvacearum causes substantial yield losses of cotton in Nigeria annually. A field study was carried out for two seasons to investigate the reaction of a selection of 10 different cotton genotypes to bacterial blight and to examine the relationship between the area under disease progress curve (AUDPC) and the yield of seed cotton in the northeastern cotton region of Nigeria. All 10 genotypes reacted positively to the disease with SAMCOT-13 having the highest incidence particularly at Dowaya and Kem locations during the two seasons. This investigation also revealed that SAMCOT-13 and Ex-Benin had the highest AUDPC while TX-CDP37HH-1-83, TAMCOT CAMD-E and TAMCOT SP-21S had the lowest AUDPC indicating relative susceptibility and resistance, respectively. However, it was observed that TX-CDP37HH-1-83, SAMCOT-11 and Ex-Benin had the highest mean yields of 1441.5 kg/ha and 1494.7 kg/ha at Dowaya, Kem and Ngurore, respectively. The results also show a highly significant (P = 0.01) negative correlation between AUDPC and yield in the three locations. These findings suggest that TX-CDP37HH-1-83, TAMCOT CAMD-E and TAMCOT SP-21S are moderately resistant to the disease in view of their lower AUDPC and are considered promising.  相似文献

Fusarium langsethiae is a toxigenic fungal species that has been reported in European small‐grain cereal crops such as oats, wheat and barley. Although its relative contribution to fusarium head blight (FHB) symptoms is not well understood, it is reported to contaminate these cereals with high levels of HT‐2 and T‐2 trichothecenes mycotoxins that are currently under consideration for legislation by the European Commission. Ten commercial oat fields in Shropshire and Staffordshire (two adjacent counties in the Midlands) in the UK were surveyed in the 2006/2007 growing season. Samples were taken from predetermined field locations at Zadoks growth stages 32/33, 69, 77‐85 and 90‐92 for F. langsethiae biomass and HT‐2 and T‐2 toxins quantification. The results from this study showed that oats can be heavily infected with F. langsethiae and have high concentrations of HT‐2 and T‐2 toxins with no apparent FHB symptoms. The regression of HT‐2 + T‐2 toxins on F. langsethiae DNA concentration was highly significant (P < 0.001, r2 = 0.55). The results indicated that although F. langsethiae had no direct effect on crop yield, it may result in indirect economic losses where the grain can be rejected or downgraded as a result of intolerable levels of HT‐2 and T‐2 toxins, which are of human food and animal feed safety concern. The influence of cultural field practices on the infection and HT‐2 and T‐2 toxins accumulation in oats was not clear and warrants further studies to identify the sources of F. langsethiae inoculum and conditions favourable for infection and mycotoxin production.  相似文献

为了探讨水稻白叶枯病成株抗性是否与防卫基因相关 ,通过RT PCR研究了几种防卫基因PAL、LOX、PBZ1、Cht 1和PR1a的表达 ,同时也分析了PAL和LOX的酶活性变化 .结果表明 ,苗期PAL受病原菌诱导上调表达 ,而成株期病原菌和伤害均可诱导PAL上调表达 ,且成株期诱导表达强于苗期 .LOX在苗期和成株期都可受病原菌诱导表达 ,但成株期表达强于苗期且更持久 .PBZ1在苗期和成株期均受病原菌和伤害诱导表达 ,但成株期表达较苗期早且更强 .PR1a和Cht1没检测到期望的扩增片段 .PAL和LOX的酶活性分析结果与基因表达分析结果一致 .因此 ,PAL、LOX和PBZ1在苗期和成株期的表达差异可能与水稻白叶枯病成株抗性相关 .  相似文献
